Walkthrough
This quest has a bug where if you accept the quest after rebirth, then switch to recruit the officer for the quest, the quest will still exist.
First, we appear at our side's Base 5 and are given the task by the Training Officer to defeat 500 enemies, as shown below:
Defeating 500 enemies shouldn't be difficult. At 250 defeats, the Training Officer will prompt: "About half a minute left." Don't worry about this message - reportedly it's a translation issue. In the Japanese version, the original meaning should be "half remaining," meaning half the defeat count remains.
After defeating 500 enemies, you'll be prompted to capture 3 bases in the upper right of the map. The capture method is: Base 1 with Officer, Base 2 with Soldier, Base 3 with Captain. The marsh map has two barriers that everyone needs to break through forcefully...
The soldiers or officers in the 3 bases have high health. There are some seal chests outside the bases, so don't be stingy with Musou... Use Musou directly to resolve Captain and Officer bases.

After capturing the base, the Training Officer will prompt again to defeat 5 Guards (some have HA super armor). At the start, you can save a 2x multiplier and lure two enemies together for a Musou kill. (My twin swords are TX brand unmodified twin swords... unfortunately... still need to grind a few more hits after Musou ends...)
There are seal chests next to the base below, which are very helpful for killing Guards. Guard 3 and Guard 5 both drop seals, use them wisely.
Quest complete, remaining 4 minutes 59 seconds or more gets S rank~~
